Essential Oil Phototoxicity Testing: IFRA Requirements for Citrus Oils

Cold-pressed citrus oils are among the most popular ingredients in fine fragrance, candles, and personal care, and they are also among the most regulated. Naturally occurring furocoumarins such as bergapten, psoralen, and citropten absorb UVA radiation and trigger phototoxic skin reactions. For B2B buyers, phototoxicity compliance is not optional: IFRA Standards restrict these materials in leave-on products, and the EU Cosmetics Regulation imposes its own limits.

Why Citrus Oils Are Different

Steam-distilled citrus oils are generally safe because furocoumarins are non-volatile and remain in the still residue. Cold-pressed (expression) oils retain them. The highest-risk materials are:

  • Bergamot oil expressed (Citrus bergamia) — historically the worst offender, with bergapten levels of 0.3 to 0.5 percent in untreated oil.
  • Lime oil expressed (Citrus aurantifolia).
  • Lemon oil expressed (Citrus limon).
  • Bitter orange oil expressed (Citrus aurantium).
  • Grapefruit oil expressed (Citrus paradisi).
  • Rue oil and fig leaf absolute, which are restricted outright in many categories.

The Reference Test: 3T3 NRU PT

The internationally accepted in vitro method is OECD TG 432 — In Vitro 3T3 NRU Phototoxicity Test. It replaced animal phototoxicity testing in the EU and is referenced by IFRA, SCCS, and most national regulators.

The protocol:

  1. Murine 3T3 fibroblasts are exposed to the test material in two parallel plates.
  2. One plate is irradiated with a defined UVA dose, typically 5 J/cm2.
  3. The other plate is kept in the dark as a non-irradiated control.
  4. After 24 hours, cell viability is measured by Neutral Red Uptake (NRU).
  5. The Photo-Irritation Factor (PIF) and Mean Photo-Effect (MPE) are calculated from the dose-response curves.

Pass/Fail Criteria

A “predicted phototoxic” result triggers use restrictions, additional in vivo confirmation, or outright exclusion depending on the customer standard.

IFRA Limits Buyers Should Know

IFRA Standards set maximum use levels for phototoxic materials in finished products. Representative limits in the current IFRA Standard include:

  • Bergamot oil expressed: restricted by bergapten content; the oil must typically be used so that bergapten in the finished consumer product does not exceed 0.0015 percent (15 ppm) for leave-on products.
  • Lime oil expressed: restricted, with low-bergapten (“lime terpenes” or distilled lime) grades preferred.
  • Bitter orange leaf (petitgrain) and bitter orange peel expressed: category-specific limits.
  • Rue oil (Ruta graveolens): effectively prohibited in fragrance use.

IFRA also publishes a positive list of non-phototoxic citrus grades, including bergamot FCF (furocoumarin-free), lime distilled, and lemon distilled, which can be used without phototoxicity restriction.

Additional Analytical Verification

The 3T3 NRU PT is a biological screen. Buyers should also require chemical quantification of the relevant furocoumarins by HPLC-UV or LC-MS/MS, with reporting in mg/kg:

  • Bergapten (5-methoxypsoralen)
  • Psoralen
  • Citropten (6,7-dimethoxycoumarin)
  • Bergamottin
  • Epoxybergamottin

A typical buyer specification for cold-pressed bergamot oil used in leave-on products is bergapten below 15 ppm in the finished product, with the oil itself often supplied as bergapten-reduced (below 0.01 percent) or FCF grade.

Documentation Buyers Should Require

A defensible phototoxicity file for a citrus oil lot includes:

  • 3T3 NRU PT report per OECD TG 432, with PIF and MPE values.
  • HPLC furocoumarin profile with quantification of bergapten, psoralen, and citropten.
  • IFRA Certificate of Compliance stating the maximum use level by product category.
  • GC-MS fingerprint confirming the oil is the declared botanical and process (expressed vs distilled).
  • Allergen declaration including citral, limonene, linalool, and geraniol where relevant.

Common Failure Modes

  • Substituting expressed oil where the formula assumed distilled oil.
  • Using bergamot oil without verifying bergapten content.
  • Ignoring phototoxicity for “natural” marketing claims, which actually increase risk.
  • Testing the neat oil but not the finished product, where synergistic effects can occur.

Closing Note

Phototoxicity is one of the few safety issues where a single analytical number can determine whether a product can legally launch in the EU. Buyers who require both 3T3 NRU PT data and HPLC furocoumarin quantification, and who default to FCF or distilled citrus grades for leave-on products, eliminate the entire risk class.
